Good part. The gBlock provided by IDT was perfect and the protocol provided on the main page worked after a couple of trial runs. We were able to make sRNA fusions to target expression of the fliC gene from E. coli (BBa_K1963004 and BBa_K1963010). The fliC target, encoding flagellin, was chosen as a Proof-of-Concept, since successful sRNA inhibition should affect swimming, which can be tested in the laboratory, but the flagellum is also important for pathogenesis in many bacteria.
